SUMMARY:
Among the natural products most used in traditional medicine, oleoresin from Copaifera reticulata Ducke (Copaíba) stands out as a rich source of bioactive compounds, especially sesquiterpenes, known for their anti-inflammatory properties. However, the precise mechanisms by which oleoresin exerts its anti-inflammatory action have not yet been completely clarified. Furthermore, there is an open question regarding the optimal formulation and dosage to ensure an effective and safe anti-inflammatory effect. Therefore, this study has the general objective of evaluating how oleoresin from Copaifera reticulata modulates the production of reactive oxygen species (ROS) by neutrophils in human blood. To achieve this goal, we propose a series of specific objectives, including the evaluation of the physicochemical characteristics of oleoresin, its cytotoxicity on neutrophils, its antioxidant capacity in a cell-free system, and its effect on ROS production by neutrophils. This is an in vitro analysis method, which includes carrying out cytotoxicity tests, evaluating the antioxidant activity of oleoresin and an assay to evaluate its effect on the production of ROS by neutrophils, using luminol-dependent chemiluminescence with different concentrations. oleoresin and standardized times. It is noteworthy that neutrophils will be obtained from human peripheral blood samples from healthy volunteer participants, aged between 20 and 40 years and equally distributed between both sexes. Therefore, the results are expected to demonstrate the ability of oleoresin to modulate neutrophil activity during inflammatory processes, inhibiting the generation of reactive oxygen species. This could position Copaíba oleoresin as a promising natural product for the development of herbal medicines with anti-inflammatory properties, since this study aims to contribute to the advancement of knowledge about the effects of Copaíba oleoresin in regulating the inflammatory response mediated by neutrophils.
