The growing interest in new natural antimicrobial agents has led to research into littleexplored organisms, such as the endophytic fungi of medicinal plants. These fungi establish a symbiotic relationship with plants, producing secondary metabolites that help plants adapt and defend themselves against biotic and abiotic stresses. Protium heptaphyllum was chosen for study due to its wide distribution in the Amazon, use in traditional medicine and morphological characteristics that favour colonization by endophytic fungi. The aim of this research is to isolate and characterize endophytic fungi isolated from Protium heptaphyllum leaves and to carry out a chemical and antimicrobial evaluation of their organic extracts. The samples of P. heptaphyllum were collected on Ufopa's Tapajós campus in Santarém-PA and identified by means of an exsiccata deposited in the Ufopa Herbarium. For cultivation, the material was first disinfected and then the leaf fragments were placed in Potato Dextrose Agar (BDA) with amoxycillin for fungal isolation, incubated at 28°C and purified. The isolates were preserved in test tubes with BDA and distilled water using the Castellani method. Identification included macroscopic and microscopic characterization, DNA extraction, PCR amplification and sequencing. For the production of fungal extracts, the fungi will be cultivated on rice and extracted with methanol, hexane and ethanol. The chromatographic profile will be analyzed by CCD, and the antioxidant activity will be evaluated by the DPPH method, in addition to the quantification of total phenolics and determination of the Minimum Inhibitory Concentration (MIC) for antimicrobial analysis. Descriptive statistics (means, standard deviations), normality tests (Shapiro-Wilk), Student's t-tests for comparisons between two groups or ANOVA for more than two groups, and Pearson's or Spearman's correlation tests to assess relationships between variables will be used to analyze the data. In view of this, it is hoped to obtain a collection of endophytic fungi from samples of P. heptaphyllum plant tissue, identified and characterized morphologically and molecularly. Secondary metabolites will then be extracted and characterized from the fungal biomass. The extracts will be tested for antimicrobial activity against pathogenic microorganisms, including gram-positive and gram-negative bacteria. The results will contribute to the understanding of the Amazonian microbiota and the antimicrobial potential of the metabolites produced by the endophytic fungi of P. heptaphyllum.
