RESUMO:

Anxiety and depression disorders have affected about 300 million people worldwide, the
conventional pharmacological treatments available have several side effects and limited
efficacy. Thus, the search for new therapeutic agents derived from plants and their constituents
have been pointed as promising sources for the treatment of disorders of the central nervous
system. Therefore, the objective of this study was to evaluate the anxiolytic and antidepressant
potential by
Myrcia sylvatica essential oil inhalation in animals, as well as to verify their
interference in motor coordination, memory and learning. For this, male mice, weighing
between 35-45 grams were used. To evaluate the anxiolytic and antidepressant activity, the
animals were divided into five groups, positive control group (anxiolytic - diazepam 5 mg/kg,
oral / antidepressant - imipramine 15mg/kg, oral), negative control group (hydroalcoholic
solution - 20%, inhalation) and three experimental groups treated with
M. sylvatica essential oil
(0.1, 1 and 2%, inhalation). For the administration of the essential oil an inhalation apparatus
was used, in which the animals were submitted for five minutes and then submitted to the
elevated plus maze (EPM) and light-dark box (LDB) to evaluate the anxiolytic activity; Tail
suspension (TS) and forced swimming (FS) to evaluate the anti-depressive activity and to
evaluate the action of the essential oil in the motor coordination, memory and learning the
animals were submitted to rota-rod, Y-maze and morris water maze tests. The results show that
in the EPM test, inhalation of
M. sylvatica essential oil at doses 0,1 and 1% significantly
increased the number of visits and the time spent in the open arms. In the LDB test, the 1%
dose of essential oil significantly increased the permanence time in the light box. In the TS and
FS tests at doses 0.1 and 1% reduced immobility time. In rota-rod, Y-maze and Morris water
maze tests no significant difference was observed between groups. These results indicate that
the essential oil of
M. sylvatica has potential in reducing the rate of anxiety and depression, not
interfering in memory, learning and motor coordination of the animals, being an indicative for
its use in aromatherapy. Consequently, other tests are necessary to know the possible
mechanisms of action of the same.
