RESUMO:

Cassava (M. esculenta) is the fifth most important crop in the world playing an
important role in human and animal feeding, and having a fundamental
socioeconomic role for family agriculture in the Amazon. Northern region leads the
ranking of the largest producer with 38% of the national production, and Pará leads
among the states with 4.2 million tons. However, this crop has been have serious
problems with root rot caused by different phytopathogens, where the main one
belongs to
Fusarium genus. In the western region of Pará, the rot of cassava roots
has limited production in the communities of the different counties. The purpouse will
be a survey and fungi characterization associated to root rot of cassava in the
western region of Pará, as well as the study of the genetic variability of the
Fusarium
solani
complex. Roots with roots rot symptoms will be collected in Santarém, Juruti,
Monte Alegre, Itaituba, Belterra and Mojuí dos Campos counties, and taken to the
Laboratory of Genetics of Interaction (LGI) for isolation, morphological and molecular
characterization and genetic variability study. Morphological identification will be
done through reproductive structures and consulted specialized materials. DNA
extraction will be performed by the Wizard® Genomic DNA Purification kit
(Promega). The molecular identification will be performed amplification and
sequencing of the ITS (Internal Transcribed Spacer) region. The genetic variability
study of
F. solani will be detected using the RAPD technique, using 10 primers of
arbitrary sequences. It is expected the data to provide a diagnosis of the diseases
affecting the cassava roots of the western region of Pará and identify the main
agents responsible for root rot of cassava, as well as to know the genetic variability of
F. solani.
