RESUMO:

Cassava (Manihot esculenta) has great socioeconomic importance in Brazil, mainly in the North and Northeast regions. In the western region of Pará, cassava production shows losses due to a complex of fungi that cause root rot. Correct diagnosis of the pathogen is essential to develop strategies for the management and control of these diseases. Thus, the objective of this work is to develop molecular tools for the detection of the causal agents of cassava root rot. It is proposed the development of two techniques, multiplex PCR and / or PCR-RFLP for these pathogens of the genera Fusarium, Neoscytalidium, Phytophthora, Phytopythium and Scytalidium. Isolates previously obtained in the region and identified at the UFOPA Interaction Genetics Laboratory will be used. The extraction of the genetic material will be carried out by the Wizard® Genomic DNA Purification kit (Promega). For the PCR-RFLP, the ITS region will be amplified with universal primers and then cleaved with restriction enzymes previously selected in silica, to obtain a restriction pattern for each pathogen. For the development of the multiplex PCR it will be carried out in two stages. First, the design of several pairs of specific primers for each pathogen will be carried out, validating their specificity. In the second part, the compatibility between the primer pairs will be evaluated for the simultaneous diagnosis of pathogens, the best combinations of primer pairs will be validated with root rot obtained from the field. Thus, he hoped to develop a molecular test for isolated pathogens, but which have similar cultural characteristics, and a diagnosis directly of the roots with cassava rot. These tools serve as a quick and efficient diagnosis for the diseases that affect the cassava roots both in the western region of Pará, and for others in Brazil.
