SUMMARY:

Antimicrobial PeptidesAMPscomprisea class of small peptides,containingfrom5 to100 amino acid residues, which are part of the innate immune system of living beings. These molecules represent a therapeutic potential, as they have a wide range of inhibitory effects against bacteria, fungi, parasites and viruses. The structure of AMPsis mainly made up of cationic and hydrophobic amino acids, which give them cationic and amphiphilic characteristics, enabling interaction with the cell membrane of microorganisms. Among the various sources ofAMPsare the secretions of venomous
animals, such as the venom of the Amazonian black scorpionTityus obscurusGervais, 1843. The objective of this study is to characterize the antimicrobial activity of three AMPs(COB21, COB22, COB23)previouslyisolatedfrom the venom ofT. obscurus. To obtain theAMPs, anenrichedfractionwith peptides will be subjected to reversed- phase High Performance Liquid Chromatography (RP-HPLC) using a C18 analytical column(4.6x250mm)previouslyequilibratedwithbufferA(H2O+0.12% trifluoroacetic acid-TFA). The elution of the peptides will beperformedin a linear gradient of buffer B (Acetonitrile + 0.1% TFA) and monitoring of the absorbance at 215 and 280 nm. The antimicrobial activity of the isolatedAMPswill be characterized according to theClinical Laboratory and Standards Institute (CLSI), by the Diffusion Discmethods(M02-A11andM44-
Aforfungiandbacteria,respectively)and determination of theMinimumInhibitory ConcentrationMIC(M07-A6for bacteria and M27for fungi). The hemolytic activity ofAMPswill be evaluatedonmouse erythrocytes andthe molecular masses determined byMALDI-TOF. Amino acid sequences will be
obtained byautomaticEdmandegradation technique.AMPsexhibit pharmacodynamic properties that reduce the resistance of microorganisms and this has aroused interest in using them as alternative sources of new antibiotics.
