SUMMARY:

A new approach called metabarcoding for environmental DNA has grown considerably in recent years. This method aims to identify the diversity of taxa and the composition of biodiversity. With the help of this technique, the aim of this research is to identify the diversity of Arthropods in an Amazon floodplain during the flood and drought periods of 2022. The water samples used in this work were collected in the Arapixuna district, in the municipality of Santarém-PA. There were a total of 7 collection points and at each point a 1L sample of surface water was obtained. At the same time as the samples were collected, measurements were taken of water parameters such as conductivity, temperature, dissolved oxygen, depth, total dissolved solids and turbidity. Immediately after collection, the water samples were filtered using a vacuum pump. After DNA extraction and PCR, we sequenced the Cytochrome Oxidase C subunit I (COI) gene, a region most commonly used in arthropod DNA metabarcoding studies. All the data analysis steps were carried out using the R 4.5.0 software. By using the environmental DNA technique to analyze arthropod diversity, we obtained a final list of 5,045 MOTUs. We found significant differences in the composition of the arthropod community between points and between seasons. Our dataset revealed a total of eight (08) orders and 13 families. The most diverse orders were Coleoptera, followed by Diptera and Lepidoptera, while the highest relative abundances at family level belonged to Cecidomyiidae, Carabidae, Cerambycidae, Chrysopidae and Palaemonidae. Environmental changes over the years can be catastrophic, producing years that can alternate between extreme droughts and floods, thus drastically affecting the species that appear year after year. Thus, these results offer a non-invasive, sensitive and effective way of understanding and helping to monitor arthropod diversity in floodplain areas. In addition, it serves as a bridge between scientific research and the practical conservation of this ecosystem.
