RESUMO:

Inflammation is a series of protective and regenerative responses of the human body, being characterized by a series of events where the inflammatory mediators like cytokines and cells are involved, being classified in acute or chronic inflammation. In this process are involved leukocyte migration, recognition of PAMPs and DAMPs by TLR receptors by tissue macrophages, production of nitric oxide, cytokines and eicosanoids such as PGE2. Non-steroidal anti-inflammatory drugs (NSAIDs) are the most consumed and their action is due to the mechanism of inhibition of COX enzymes, whereas glucocorticoids are drugs that have anti-inflammatory effects with an action mechanism associated with the binding with the cytosolic glucocorticoid receptors , regulating the gene expression of immune cells. About salicylates, they have anti-inflammatory effects, as well as their derivatives, however, they have a series of adverse reactions, which include gastrointestinal, renal and cardiovascular problems. The creation of new molecules has become an art for scholars of the area, because of the need related pharmaceutical industries, techniques such as this are important for the development of new drugs such as those derived from salicylates. NSAIDs have proven toxicity and are considered to be harmful to the renal and gastrointestinal and cardiovascular systems. Thus, methyl salicylate derivatives with their anti-inflammatory effects are an option in the development of new drugs to be used to combat inflammation. The objective of this work is to design, synthesize and evaluate the anti-inflammatory activity of Methyl Nitro-Salicylate in vitro in macrophages. To obtain the SMN, we will start from a reaction performed between three compounds, the assembly and production of this new drug were carried out in partnership with the Laboratory of Pharmaceutical Chemistry of the Federal University of Pará. Cell viability will be performed by MTT method, NO production by the Griess method, the dosages of IL-1β, TNF-α and PGE-2 will be quantified by the ELISA method. As preliminary results we present the reaction performed between some compounds such as methyl salicylate and nitric acid in medium containing strong sulfuric acid, which resulted in the formation of Methyl Nitro-Salicylate. Regarding cell viability, the preliminary results showed that SMN did not promote a cytotoxic effect at concentrations below 300 μM after 24 h of incubation, causing cell death at the concentration of 1000 μM, in which a significant decrease in cell viability was observed.
