SUMMARY:

Malaria is an infectious and parasitic disease that affects millions of people in tropical and subtropical regions of the world. In Brazil, most are in the Legal Amazon, among other factors, due to the conditions required for the existence of the vector. Malaria, a disease of the protozoan genus Plasmodium, is a major impacting protozoan in the world. Plasmodium falciparum resistance to artemisinin presentation in Southeast Asia or malaria control. There is recognized importance of natural products, including plant products, in the development of modern therapies. Medicines or natural diseases are able to treat about 80% of the diseases that humanity has. Libidibia ferrea Mart. ex Tul. var. ferrea, is a tree that grows throughout Brazil, being found mainly in the North and Northeast regions, being commonly known as jucá. Jucá (Libidibia ferrea) has been extensively investigated from the point of view of its various biological properties such as: anti-inflammatory, analgesic, anticancer, anti-ulcer, antioxidant and antimicrobial. In this context, the present dissertation sought to evaluate the in vitro anti-plasmodial activity of the hydroalcoholic extract of Libidibia ferrea Mart. ex Tul. var. ferrea (EHJ). The preparation of the hydroalcoholic extract of Libidibia ferrea was carried out according to AMÉRICO et al. (2020) adapted by KOBAYASHI et al., (2015). A human cell line used for toxicity analysis of WI-26VA4 (ATCC CCL-75 lung fibroblast) cultured by the method described by PEREIRA et al. (2012). For the cytotoxicity assay, the MTT method was used. This test consists of the rapid colorimetric evaluation of the growth of cell lines in vitro. The assay is based on the reduction of the tetrazolate salt by the enzyme hydrogenase succinate present in the mitochondria of viable eukaryotic cells, forming a water-insoluble crystal known as a formazan crystal. When solubilizing the crystal with an organic solvent, it acquires a violet configuration that is spectrophotometrically. To complement the intracytocytic phases of Plasmodium falciparum, paras of the W2 strain were cultured in human red blood cells in vitro under conditions conditions by TRAGER & JENSEN (1976). For the plasma test, P. falciparum cultures with 0.5% ring-stage parasitaemia and 2% hematocrit were distributed in a 96-well plate (180μL per well). The compound was added (20μL) to the test plate, in triplicate, and in different series of 50 μg/mL. In tests performed, cytotoxicity was recorded as the reduction in absorption. The Jucá hydroalcoholic extract did not show cytotoxic activity with an IC50 value greater than 100 µg/ml. In determining the IC50 by anti-plasmodial test, the value was 11.10 µg/ml. In addition, the selectivity index (SI) 9 was obtained, which corresponds to the relationship between the cytotoxic and antiplasmodial activities of the hydroalcoholic extract of Jucá. The extract did not have hemolytic activity in any of the presentations presented. In his possession, the results may determine that the Jucá extract is a great candidate for alternative therapies in the treatment of malaria. It may also carry out studies to reinforce the findings in this study.
