SUMMARY:

Propolis has aroused great interest in the scientific community due to its pharmacological properties and safety profile, making it a possible candidate for the development of new antimicrobial drugs. Hydroalcoholic, ethanolic and aqueous extracts of propolis have exhibited remarkable therapeutic effects, including antioxidant, anti-inflammatory, antiviral, antiparasitic, antifungal, antibacterial, immunostimulant, anesthetic, healing, hepatoprotective and antineoplastic properties. As a result, propolis has great therapeutic potential in the antibacterial aspect, which has been empirically known for generations, and, in conjunction with advanced biotechnological techniques, it can be a valuable resource in the treatment of various infections. The main objective of this research is to evaluate the antimicrobial activity and carry out the physical-chemical characterization of propolis from stingless bees of different origins and species from the Brazilian Amazon region. The samples will come from donations from producers in the region and from the meliponary of the Federal University of Western Pará (UFOPA). Crude propolis will be analyzed microbiologically, and the isolates obtained will be processed through biochemical tests to identify the genera to which they belong. The molecular identification of isolates will be performed from bacterial DNA, and sequencing of 16S regions, and analysis of sequence alignments. Ethanol extracts will be obtained from a 15.00 g sample suspended in 50 mL of ethanol and stirred for 48 hours, then filtered and centrifuged. The extracts will be dried and resuspended in 80% ethanol. Aqueous extracts will be obtained in the same way, using deionized water. Geopropolis extracts will be obtained from a 45 g sample, using ethanol in a Soxhlet extractor for 6 hours, and the solvent will be evaporated and resuspended in 95% ethanol. The process will be repeated three times for each sample. The analysis of antimicrobial activity will be performed against strains of Staphylococcus aureus, Escherichia coli and Candida spp. The physical-chemical analysis of the obtained extracts will be made to determine the phenolic compounds, the determination of the total flavonoids content, and the antioxidant activity (DPPH %). It is expected that the ethanolic and aqueous extracts of propolis from native bees present bacteriostatic and bactericidal activity against the selected strains, in addition to physicochemically characterizing the obtained extracts
