SUMMARY:

Endophytes are organisms present inside plant tissues without causing apparent damage, for at least one life cycle, can alter the biochemical processes involved in endophyte-host plant relationships, producing new biomolecules, with biotechnological potential and use in different areas of the pharmaceutical and agricultural industry. The species Piper marginatum JACQ. (Piperaceae) popularly known as pimenta-do-mato, malvarisco, capeba-cheirosa is widely used in folk medicine for presenting a high therapeutic potential in the treatment of skin lesions, antimicrobial and antiparasitic activity, acaricide, insecticide, among others. Even with several ethnopharmacological uses, there are few studies that seek to study the endophytic community associated with this plant.  In this sense, the objective of this work was to evaluate the diversity of endophytic fungi associated with P. marginatum (Piperaceae). Leaf fragments were superficially disinfected in 70% alcohol, in 3% hypochlorite and again in 70% alcohol, sterile distilled water and inoculated on Potato Dextrose Agar (BDA) culture medium for isolation of endophytic fungi. The plates were incubated for up to 30 days and the fungi obtained isolated and purified on BDA. All fungi obtained were preserved and will be deposited in the Collection of Microorganisms of the Federal University of Western Pará. The fungal colonization rate was evaluated by macroscopic observation and grouped according to morphological characteristics. The identification of fungi was performed by sequencing the internal transcribed spacer (ITS1and ITS4) of the rRNA gene. For fungal DNA extraction, a method using the WizardÒ Genomic DNA Purification kit, Promega, was used. The DNA sequences of the endophytic fungi will be compared with the sequences, deposited in GenBank using the BLAST program (Basic Local Alignment Search Tool - version 2.215 of BLAST 2.0 available on the NCBI portal (http://www.ncbi.nlm.nih.gov/blast/), developed by the National Center For Biotechnology. The fungi that presented sequences with E value = 0, coverage and identity ≥99%, as well as proximity when phylogenetically analyzed with sequences of type or reference species, using the MEGA 6 program (Molecular Evolutionary Genetics Analysis).  After purification, 171 isolates of filamentous fungi were obtained, the fungal colonization rate was 98.33% which were grouped using their morphological characteristics into 18 morphogroups. In the morphological analyses, they were classified into the following genera according to the characteristics of the colonies.
