RESUMO:

Viruses are infectious particles, highly adaptable and capable of infecting different human tissues, such as SARS-CoV-2, (Severe Acute Respiratory Syndrome, Coronavirus 2) responsible for the disease COVID-19. Given the above, it is intended to monitor and identify the presence of SARS-COV-2 in wastewater at strategic points, using an analytical method, fast and accurate. The activities will be assisted by professionals from Sanitary and Environmental Engineering, Biology and Computation and Chemistry. The study will take place in the urban area of Santarém at three sampling points for wastewater collection: (i) EEE from Residencial Salvação (ii) Wet well ETE Irurá and (iii) ETE from Regional Hospital. 500 ml of samples will be collected every 12 min, for 4 h, to obtain a final composite sample of 2L. The analyzes will occur in parallel with the collections every fortnight in the period from February to September 2021, totaling 16 determinations. In the Wastewater Treatment Laboratory, the physical-chemical parameters of the samples will be analyzed (Temperature; pH; Alkalinity; Dissolved Oxygen; Phosphorus; Solids; Nitrogen, Ammonia; Chemical Demand for Oxygen, Nitrite and Nitrate) and subjected to the analytical methods of the Standard Methods for the Examination of Water and Wastewater (2012). The biological analyzes will be carried out at the Molecular Biology Laboratory where they will be submitted to the rupture process to release the virus from the intestinal and respiratory cells according to the procedure described by Jacinavicius et al. (2013), and pre-treatment (separation and filtration). The RNA extraction will follow the protocol established by the manufacturer of the All Prep Power Viral DNA / RNA kit (Qiagen®, Hilden, Germany) and the identification and quantification of SARS-CoV-2 by real-time Polymerase Chain Reaction with reverse transcription (RT-qPCR). The data will be interpreted quantitatively via the construction of analytical curves with plasmids containing the SARS-CoV-2 marker genes. The result of the detection of SARS-CoV-2 at the sampling points associated with the technical information of the sanitary sewage network, will generate a direct indicator of the existence, or appearance, of possible sources of contamination. Therefore, it is expected that the monitoring of wastewater can provide an effective approach to health and basic sanitation authorities, helping to control the spread of the virus and to develop measures to reduce the risk of contagion to the population.
