Leishmaniasis is considered a neglected endemic disease that occurs 22 species that affect humans, of which 12 occur in Brazil. The treatment conventional disease of leishmaniasis tends to be painful and invasive, resulting in therapeutic abandonment. The plant species of common occurrence in the Amazon Region Piper marginatum, has shown in vitro leishimanicidal therapeutic potential under leishmaniasis integumentary. The development of a topical drug containing P. marginatum would enable a less painful treatment in addition to generating a new alternative of drug therapies and the commercialization of a new herbal product. With this, the work aims to carry out the chemical characterization of the ethanolic extract of P. marginatum for the development and standardization of a gel formulation containing IFAV that makes it possible to evaluate whether there is leishmanicidal and cytotoxic activity of the formulations in vitro and in vivo front Leishmania amazonensis. Initially, collection was carried out in February 2023, where the moisture content of the vegetable drug that was used to prepare the extract, the extraction method was hot, via Soxhlet, phytochemical screening was carried out a thin layer chromatography (TLC) for the search for simple and glycosylated, terpenes, condensed and hydrolyzable tannins, fatty acids and flavonoids, From this, it was followed by the quantification of phenolic compounds, based on a calibration by spectrophotometric method with Folin-Ciocalteu reagent and quantification of flavonoids, using the calibration curve by spectrophotometric method with reagent aluminum chloride. The moisture content of the leaves was 6.9 ±0.2%, the qualitative investigation by CCD, demonstrated that there was a strong presence of flavonoids, mild to moderate for terpenes, condensed and hydrolyzable tannins and fatty acids. The quantitative content of phenolic compounds 16425.97± 3.08 µg.EAG.100g -1 and flavonoids 68,872.56± 13.94 µg.ERUT.100g - . Based on the results, the extract will be standardized by high performance liquid chromatography (HPLC), formulation design and screening for in vitro testing on the L. amazonensis strain later.
