SUMMARY:

Despite advances in the formulation of drugs for the treatment of skin lesions, the use of medicinal plants has become increasingly valued as they have several advantages.  Copaiba is one of the medicinal plants of considerable interest to science, since its oleoresin has several beneficial properties reported in the literature, including healing activity.  Therefore, this study aims to evaluate the healing potential of copaiba oleoresin (Copaifera reticulata Ducke) and its volatile and resinous fractions on skin wounds in Wistar rats (Rattus norvegicus albinus).  To achieve this, initially the hydrodistillation of copaiba oleoresin and the chemical characterization of its fractions will be carried out.  Next, the dermal toxicity of repeated doses of crude oleoresin, essential oil and resin will be evaluated in male and female rats for 21 days.  For this experiment, 75 male rats will be used, divided into 5 groups, with 15 animals each: negative control group (NG), in which no substance will be applied; positive control group (GD), with the application of dexamethasone; essential oil group (GOE); resin group (GR) and oleoresin group (GOR).  Thus, after creating a circular lesion of 8 mm in diameter on the back of the animals using a dermatological Punch, the wound will be treated daily topically with 0.1 ml of the substance corresponding to its group.  To evaluate the healing course of the wounds, on the third day (day 3) of treatment, five animals from each group will be chosen at random and euthanized for macroscopic and microscopic evaluation, as well as on days 12 and 21 of treatment.  The macroscopic evaluation will be by superficial measurement of the lesion area through photographs and the microscopic evaluation will be through histological analysis of the inflammatory infiltrate, blood vessels, fibroblasts and collagen.  Considering the complexity of tissue repair and the damage resulting from wounds, it is necessary to find different compounds that can contribute to accelerating the healing process.  Thus, Copaifera reticulata Ducke oleoresin and its fractions are expected to contribute to the healing of skin wounds in Wistar rats.
